Tuesday, November 24, 2009. RPA-2500 and highly sensitive dyes. Problem - customer can't see the SybrGreen in the gel. Switched from Invitrogen's ultra pure agarose to the RPA-2500. Ran side by side tests and had the issues w/ only our agarose. Using a TAE buffer that he makes and dilutes to 1x. Per Arup Sen, the RPA-2500 is meant for routine mol bio applications (such as when using EtBr). It has not been tested to work with highly sensitive dyes such as Sybr. Wednesday, August 12, 2009. GelRed Post Staining Recommendations. Our recommended concentration for post-staining is 1X final concentration. So from a 10,000X stock of GelRed, 100uL into 1 L would be the correct dilution. But it is not necessary to use 1X TAE. You can if you'd like, but just water is also ok. If you want a 3X final concentration for post-staining, you will need to add 300uL to 1 L. The correct dilution would be to have a 3X final concentration from 10,000X stock for post-staining.
